ควีนออฟเดอะเซาต์ Queen of the South Season 2 เล่าถึงเรื่องราวอันทรงพลังของ Teresa Mendoza ที่ถูกบังคับให้ต้องทำงานกับแก๊งค์ที่เพิ่งฆ่าแฟนหนุ่มของเธอ เธอจึงต้องพึ่งพาสติปัญญา เพื่อนผู้ภักดี และสมุดโน๊ตลึกลับเพื่อเอาตัวรอด เธอได้หนีมายังอเมริกาหลังจากที่แฟนหนุ่มของเธอผู้เกี่ยวข้องกับขบวนการขนเสพยาเสพติดถูกฆาตกรรมอย่างไม่คาดฝันในเม็กซิโก หลังจากนั้นเธอได้ค่อยๆสร้างทีมจากสมุดโน๊ตลึกลับจากอดีตของเธอเพื่อโค่นล้มขบวนการค้ายาเสพติดที่มีอยู่ให้หมดไป
